Pharmaceutical Water Systems

Potable Water → DM / Purified Water (PW)

Pharmaceutical water from source to point of use — the pharmacopoeial framework and its actual numbers, the generation engineering, and the storage, distribution and surface integrity that keep the water fit once it is made.

  • WHO TRS 1033
  • WHO TRS 1025

  • USP <643>
  • ISPE Baseline Guide Vol. 4
  • ASME BPE
  • IS 10500

A water system is the one utility that is also a product ingredient, and the only one whose qualification is measured in seasons rather than days.

This module covers the pharmacopoeial framework and the numbers behind it, the generation train that produces the water, and the loop, surfaces and sanitisation regime that decide whether it is still fit by the time it reaches a point of use.

What this module covers

Origins
From municipal chlorination in 1908 to the 1996 shift that replaced a dozen wet-chemistry tests with two real-time measurements.
Standards map
EPA and WHO drinking water frameworks, WHO TRS 1033 and 1025, USP and Ph. Eur. chapters, ISPE Vol. 4, ASME BPE, IS 10500.
Potable water fundamentals
Source types, coagulation, filtration and the four disinfection routes — with what each one leaves behind for the plant to deal with.
Pharmacopoeial grades
PW, WFI, Highly Purified Water and the packaged grades, and which pharmacopoeia recognises which.
The actual limits
Conductivity, TOC and microbial figures for USP and Ph. Eur. side by side — including where the two genuinely differ.
Staged conductivity
What Stage 1, 2 and 3 are for, and the two instrument and interpretation errors that cause facilities to reject good water.
Generation engineering
Pretreatment, reverse osmosis, EDI, the classic DM train, and where distillation still earns its place.
Loop design
Continuous circulation, turbulent flow rather than a headline velocity, and turnover — the primary defence against biofilm.
Dead legs
Why the familiar six-diameter rule is thirty years out of date, what replaced it, and how to assess an existing installation fairly.
Rouging & passivation
The three classes of rouge, what causes each, and why derouging without passivation leaves the surface worse than before.
Microbial control
Hot, ambient and ozonated strategies compared — and the most common misunderstanding about what UV actually does.
Storage tank design
Vent filter, spray ball coverage, drainability and relief — the four details that decide whether a tank is a barrier or a reservoir.
GEP versus GMP
Which parts of a water system are qualified and which are commissioned, item by item.
Three-phase validation
Why water systems need a year, and the two opposite mistakes facilities make with Phase 3 — both expensive.

Questions this module answers

  • Is your online conductivity instrument running in temperature-compensated mode?
  • Do you reject water on a Stage 1 excursion without running Stage 2?
  • Are your dead legs assessed against the six-diameter rule or against the current criterion?
  • Has a derouging on your site ever been followed by verified passivation?
